  • Description: This is a cancer cell vaccine consisting of autologous dendritic cells (DCs) loaded with mRNA encoding the human cytomegalovirus (CMV) matrix protein pp65 (65 kDa lower matrix phosphoprotein; UL83) as a fusion protein with the full-length lysosome-associated membrane protein (flLAMP), with potential immunostimulatory and antineoplastic activities. The vaccine exposes the immune system to the CMV pp65 peptide, which may elicit a cytotoxic T-lymphocyte (CTL) response against CMV pp65-expressing tumor cells. The incorporation of flLAMP may route CMV pp-65 antigens into the lysosomal compartment, resulting in enhanced MHC class II antigen presentation, thereby promoting CD4-positive T-cell responses. (NCIT_C156067) Granulocyte Macrophage-Colony Stimulating Factor (GM-CSF) is a powerful adjuvant capable of stimulating macrophage function, inducing proliferation and maturation of DCs, and is able to enhance T-lymphocyte stimulatory function. The vaccine can be used in combination with monoclonal antibodies, such as basiliximab, that can block tumor growth in different ways, as well as temozolomide,, and radiation therpay.
